Start the single-player campaign up as you would usually, once you take control of the character (after loading is finished) have second player press start on another controller. Doing this will enable co-op gameplay and needs to be done everytime you want to play Split-screen.
However, if you tried logging into your LIVE account from another Xbox 360 playing Resident Evil 5, you will notice that you do not have access to your character, money, weapons, etc. from the previous games saved on the previous Xbox 360 because these items are all saved on the client-side; they are not saved on the server-side.
The only way to play CO-OP is by having Xbox Live if there is another way no one has found it.
